目的:探讨IL-6抑制剂对种植体周围炎小鼠模型的疗效及其作用机制。方法:选取4周龄雄性小鼠(C57BL/6J)15只,实验总共分为三组(每组5只):对照组(植入种植体,但未进行结扎诱导)、模型组(结扎诱导,构建种植体周围炎小鼠模型)和抑制剂组(结扎诱导+腹腔注射IL-6抑制剂)。使用微计算机断层扫描(Micro-CT)检测种植体周围骨丢失,苏木精和伊红(HE)染色检测种植体周围牙龈组织的炎症浸润,使用RT-qPCR检测牙龈组织中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α) mRNA 相对表达量,使用Western blot法检测牙龈组织中NF-κB通路相关蛋白p-P65和p-IκBα蛋白表达水平。结果:Micro-CT结果显示IL-6抑制剂可减少种植体周围炎的骨丧失。HE结果显示IL-6抑制剂处理后,小鼠炎症浸润显著减少。RT-qPCR和Western blot结果显示,IL-6抑制剂可显著降低牙龈组织中IL-6、TNF-α mRNA 相对表达量以及p-P65和p-IκBα的蛋白表达水平。结论:IL-6抑制剂可抑制种植体周围炎的骨丧失和炎症浸润,这些治疗效果可能与抑制TNF-α的表达和NF-κB信号通路有关。

Objective To investigate the curative effect and mechanism of IL-6 inhibitor on mice with peri-implantitis. Methods Fifteen 4-week-old male mice (C57BL/6J) were selected, and the experiment was divided into 3 groups (5 mice in each group: the control group (implants implanted, but no ligation induction), the model group (ligation induction: construction peri-implantitis mouse model) and the inhibitor group (ligation induction + intraperitoneal injection of IL-6 inhibitor). Microcomputed tomography (Micro-CT) was used to detect peri-implant bone loss. Hematoxylin and eosin (HE) staining was used to detect inflammatory infiltration in peri-implant gingival tissue. The mRNA relative expression levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF- α) in gingival tissue were detected by RT-qPCR. The protein expression levels of NF-κB pathway related proteins p-P65 and p-IκBα in gingival tissue were detected by Western blot. Results Micro-CT results showed that IL-6 inhibitor could reduce bone loss in peri-implant inflammation. HE results showed that inflammatory infiltration in mice was significantly reduced after IL-6 inhibitor treatment. RT-qPCR and Western blot results showed that IL-6 inhibitor significantly decreased the mRNA relative expression levels of IL-6 and TNF- α, and the protein expression levels of p-P65 and p-IκBα in gingival tissue. Conclusion IL-6 inhibitors can inhibit bone loss and inflammatory infiltration in peri-implantitis, and these therapeutic effects may be related to the inhibition of TNF- α expression and NF- κB signaling pathway
