目的:探讨由苦参(S.flavescens)根、胀果甘草(G.inflata)根、黄芩(S.baicalensis)根制备而成的复合植 物提取物(SGS)对十二烷基硫酸钠(Sodium lauryl sulphate,SLS)刺激皮肤后的保护作用。方法:采用SLS或脂多糖 (Lipopolysaccharide,LPS)建立角质形成细胞、巨噬细胞、三维体外重建表皮模型、鸡胚绒毛尿囊膜(CAM)刺激反应模 型,并从细胞活力、细胞因子、基因表达、组织水平、血管反应维度分析其可能的调节机制。结果:SLS刺激后,角质形成 细胞的细胞活力下降,角蛋白10、角蛋白16、水通道蛋白3的基因表达量降低,前列腺素E2表达量增加;表皮组织活力和结 构完整性下降,细胞因子表达量增加;CAM血管损伤性出血增加。LPS作用于巨噬细胞,细胞因子表达量也显著升高。同时使 用一定浓度的SGS后,可显著提高SLS刺激损伤状态下的细胞活力、组织结构完整性,降低细胞因子的表达量,加速恢复皮肤 屏障相关基因表达水平,并缓解血管出血反应。对于LPS诱导的巨噬细胞白介素6和前列腺素E2表达量,0.003%~0.3%浓度的 SGS也具有显著性下调作用。结论:SGS对SLS或LPS诱导的皮肤细胞刺激性反应具有明显的保护作用,其作用机制包括降低外 界刺激对细胞活力和组织结构的损伤、减少细胞因子的分泌、修复皮肤屏障和缓解血管性出血反应。

Objective To study the protective eff ect of a multi-herb extraction (SGS), extracted from Sophora fl avescens root, Glycyrrhiza infl ata root, and Scutellaria baicalensis root, on skin irritation induced by sodium dodecyl sulfate (SLS). Methods SLS or lipopolysaccharide (LPS) was used to establish the study models on keratinocytes, macrophages, 3D reconstructed epidermis models (Episkin), and chick chorioallantoic membrane (CAM) from cell viability, cytokines, gene expression, tissue level and vascular response. Results After SLS stimulation, the cell viability and the level of keratin 10, keratin 16, and aquaporin 3 decreased on keratinocytes, while the expression of prostaglandin E2 increased. And also the viability and structural integrity of Episkin decreased with an increase expression of cytokines. Increased vascular injury bleeding in the CAM after SLS stimulation. LPS acts on macrophages, and the expression of cytokines is also signifi cantly increased. When treated with a certain concentration of SGS and SLS, the cell viability and tissue structural integrity was signifi cantly improved, with a reduction expression of cytokines, accelerated recovery of skin barrier-related genes and vascular hemorrhage responses. For the LPS-induced expression of interleukin-6 and prostaglandin E2 on macrophage, SGS also had a signifi cant down-regulation eff ect at a concentration of 0.003%-0.3%. Conclusion SGS has obvious protective eff ect on the irritant response of skin cells induced by SLS or LPS through reducing the damage to cell viability and tissue structure, reducing the secretion of cytokines, repairing the skin barrier and relieving vascular hemorrhage.
