摘要:: 目的：通过蛋白芯片技术筛选血清中的2个结果蛋白做免疫组化(Immunohistocry，IH)研究，检测其在干燥综合征 ( Sj?gren’s syndrome ，SS)患者唇腺组织、系统性红斑狼疮（Systemic lupus erythematosus，SLE）患者皮损及狼疮性肾炎（Lupus nephritis，LN）患者肾脏组织中的表达情况。方法：收集2004年1月-2013年12月就诊于风湿免疫科患者的组织蜡块43例，按照不同疾病、不同部位将组织蜡块分为SS组16例、非SS对照组10例、SLE皮损组9例、LN组8例。采用免疫组织化学染色法检测骨桥蛋白和铁调素在SS患者、SLE患者皮损和LN患者肾脏组织中的表达。使用Fromowitz的综合计分法进行结果判定。阳性率的比较采用四格表确切概率法。结果：骨桥蛋白和铁调素在SS唇腺组织的表达均高于非SS唇腺组织，两种蛋白在SLE皮损中未液化的少量基底层细胞中见阳性表达、狼疮肾组织中肾小管上皮细胞见阳性表达。结论：骨桥蛋白与铁调素可在干燥综合征患者唇腺组织、系统性红斑狼疮患者皮损及狼疮性肾炎患者的肾小管中表达。

Abstract:: Objective The immunohistochemical method was applied to text two proteins screened through SELDI-TOF-MS from serum. To test the expression in the labial gland tissue of SS patients,the skin lesion of Systemic Lupus Erythematosus and renal tissue in LN patients. Methods Collection forty-three tissue wax blocks from the patients who treated in the department of rheumatology and immunology during January 2004 to December 2013. The tissue wax blocks were divided into SS group(n=16), non-SS control group(n=10), SLE skin lesion group(n= 9)and LN group(n=8) accord diff erent disease and diff erent position.Used the immunohistochemical staining to test the expression in the labial gland tissue of SS patients, the skin lesion of Systemic Lupus Erythematosus and renal tissue in LN patients.Used the Fromowitz comprehensive scoring method to assess the results. The comparison of two kinds of protein staining positive rate used the four table exact probability. Results Osteopontin and hepcidin in the expression of SS labial gland tissue were higher than non-SS labial gland tissue, was detected in the lesions of SLE did see positive expression in a small amount of liquid base layer cells, lupus kidney tissues in renal tubular did see positive expression. Conclusion Osteopontin and hepcidin were expressed in labial gland tissue of SS, skin lesion of SLE and lupus kidney patients’renal tubules of LN.
