摘要:

[摘要]目的：研究甲基莲心碱对增生性瘢痕成纤维细胞中Bcl-2、Bax及Caspase-3 中mRNA和蛋白表达的影响。方法：体外
培养增生性瘢痕成纤维细胞及正常皮肤组织，随机分为4组：对照组、甲基莲心碱低、中、高剂量组(5、10、20μg/ml)。
各组分别加入不同浓度药物培养24h后，采用RT-PCR检测Bcl-2、Bax及Caspase-3 mRNA的表达，并运用Western Blot检测
Bcl-2、Bax及Caspase-3蛋白的表达。结果：与对照组相比，低剂量组Bcl-2、Bax及Caspase-3 mRNA表达量均无明显变化
（P ＞0.05）；中、高剂量组Bcl-2 mRNA表达量明显降低，Bax及Caspase-3 mRNA表达量明显增加，差异有统计学意义（P
＜0.05）。与对照组相比，低剂量组Bcl-2蛋白表达量无明显变化，中、高剂量组Bcl-2蛋白表达量明显降低，差异有统计学
意义（P ＜0.05）；低、中、高剂量组Bax及Caspase-3蛋白表达量均明显增加，差异有统计学意义（P ＜0.05）。结论：甲基
莲心碱能上调增生性瘢痕成纤维细胞中Bax、Caspase-3的表达，同时下调Bcl-2的表达。
Abstract:
Abstract: Objective To research the influence of neferine on the expression of mRNA and protein in Bcl-2, Bax and Caspase-3
in hypertrophic scar fibroblast. Methods Hypertrophic scar fibroblasts and normal skin tissue were cultured in vivo and randomly
divided into four groups, namely, the control group, low, medium and high dose groups of neferine(5,10,20μg/ml). After drugs
with different concentrations are respectively added into each group for culture for 24 hours, the expression of Bcl-2, Bax and
Caspase-3 mRNA was detected by RT-PCR, and the expression of Bcl-2, Bax and Caspase-3 protein was detected by Western
Blot. Results Compared with the control group, there was no significant change in the expression of Bcl-2, Bax and Caspase-3
mRNA in the low dose group(P ＞0.05). The expression of Bcl-2 in medium and high dose group decreased obviously, and the
expression of Bax and Caspase-3 significantly increased(P ＜0.05). Compared with the control group, there was no significant
change in the expression of Bcl-2 protein in the low dose group, and the expression of Bcl-2 in the middle and high dose groups
decreased significantly(P ＜0.05). And the expression of Bax and Caspase-3 in the low, middle and high dose groups increased
obviously(P ＜0.05). Conclusion Neferine can upregulate the expression of Bax and Caspase-3 in hypertrophic scar fibroblast
and downregulate the expression of Bcl-2 therein as well.
