[摘要]目的:探讨在-80℃低温条件下冻存不同时间的脂肪组织颗粒的结构及生物学活性变化。方法:将吸脂后的脂肪经过洗
涤、离心后低温保存,1周、4周、8周、12周、16周、20周、24周后,对复温后的脂肪组织颗粒进行大体观察、组织学观察、
活性检测、SVF细胞培养等。结果:随冻存时间的延长,脂肪组织的结构及生物学活性逐步下降,冻存1周的脂肪组织活细胞
面积(35.35±4.05)%较新鲜脂肪(79.25±7.44)%显著减少,差异有统计学意义(P <0.01),冻存1周的脂肪细胞较新鲜
脂肪细胞线粒体活性显著下降(P <0.01),SVF细胞培养,冻存脂肪来源的贴壁细胞数量较新鲜脂肪显著减少(P <0.01),
且细胞生长缓慢、状态较差。结论:大多数脂肪细胞在一次冷冻保存过程中便失活,而冻存时间的延长与脂肪细胞活性降低
的相关性并不明显,不推荐临床使用不添加保护剂直接冻存的脂肪进行移植。

Abstract:
Abstract: Objective The aim of this study was to observe the changes of histological structure and biological activity of
adipose granules frozen at -80℃ for different time. Methods The adipose tissue obtained by liposuction was stored at low
temperature after washing and centrifugation. After 1 week, 4 weeks, 8 weeks, 12 weeks, 16 weeks, 20 weeks and 24 weeks,
the granules of adipose tissue after rewarming were observed by gross observation, histological observation, activity detection,
SVF cell culture and so on. Results With the prolongation of freezing time, the histological structure and biological activity of
fat gradually decreased. The area of living cells in adipose tissue frozen for 1 week (35.35±4.05)% was significantly lower than
that of fresh adipose tissue (79.25±7.44)%, the difference was statistically significant(P <0.01). The mitochondrial activity of
adipocytes frozen for 1 week was significantly lower than that of fresh adipocytes (P <0.01). In SVF cell culture, the number of
adherent cells derived from cryopreserved fat was significantly lower than that of fresh fat (P <0.01), and the cells grew slowly
and in poor condition. Conclusion Most adipocytes are inactivated during a single cryopreservation. There was no significant
correlation between the prolongation of freezing time and the decrease of adipocyte activity. It is not recommended to transplant
directly frozen fat without protective agent in clinic.
