摘要:
[摘要]目的:建立SD大鼠实验性牙周炎动物模型,观察黄芩提取物对其IL-1β和牙槽骨骨密度等的影响。方法:建模组(牙
周炎组和黄芩提取物组)在大鼠上颌第二磨牙腭侧局部注射牙龈卟啉单胞菌液(P.g.W83),丝线结扎牙颈部,再局部涂抹
菌液,饲以软食;8周后采用50mg/L黄芩提取物局部给予药物治疗,4周后检测龈沟液和唾液中的IL-1β含量及牙槽骨骨密度
等变化。结果:8周后建模组大鼠双侧上颌第二磨牙腭侧牙龈组织红肿,易出血,牙周袋约3~4mm;龈沟液和唾液中IL-1β
明显升高;Micro CT显示牙槽骨吸收达根1/2处,根分叉区可见牙槽骨吸收低密度投射区,牙周炎模型建立成功;50mg/L黄
芩提取物治疗4周后,龈沟液和唾液中的IL-1β明显下降,牙槽骨密度、高度等显著升高。 结论:该方法能够快速、稳定、
有效地建立SD大鼠实验性牙周炎模型,50mg/L黄芩提取物能明显改善大鼠牙周炎组织中IL-1β和牙槽骨状况,为牙周病的临
床防治提供实验依据。

Abstract:
Abstract: Objective To establish an experimental periodontitis model in SD rats rapidly, and determine the effects of
Scutellariabaicalensis extracts on IL-1βand alveolar bone in SD rats. Methods After ligated the cervical region using wire
ligatures, the male SD rats in experimental group were received a direct injection of P.g.W83 into the palatal gingival tissue
of the bilateral maxillary second molar. Then the P.g.W83 bacterial solution were applied locally and feed with soft food 8
weeks. The content of IL-1 β in gingival crevicular fluid and saliva and the density of alveolar bone were measured after 4
weeks using 50mg/L Scutellariabaicalensis extracts. Results 8 weeks later, the gingival of SD rats in experimental group were
red and swollen, bleeding on probe and probing depth was about 3-4mm.The levels of IL-1β both in gingivalcrevicular fluid
and saliva were higher than control. The micro CT demonstrated the level of alveolar bone was destroyed to the half of teeth
root of the second molar tooth. Alveolar bone resorption can be seen in the root bifurcated area and buccolingual sections.
Theseindicated that the periodontitis model in SD rats was established successfully. After 4 weeks of treatment with 50mg/L
Scutellariabaicalensis extract, the level of IL-1β in decreased significantly, and the density and height of alveolar bone increased
significantly. Conclusion This method can be used to establish an experimental periodontal in SD rats rapidly, and 50 mg/
L Scutellariabaicalensis extract can significantly decrease the level of IL-1β and improve alveolar bone in periodontitis.It can
provide a basis for further study on etiology,prevention and treatment of periodontitis.
