目的:基于p38 MAPK通路探究骨髓基质干细胞对大鼠皮肤创伤修复的干预效果。方法:选取30只Wistar雄性大鼠,随机抽取10只作为正常组,剩余20只建立大鼠皮肤创伤模型,最终19只建模成功,随机分为创伤组9只,骨髓基质干细胞组10只。建模完成后骨髓基质干细胞组大鼠在创伤部位注射骨髓基质干细胞,正常组、创伤组大鼠不做处理,观察各组大鼠的变化。对比各组大鼠创口愈合率、再生上皮厚度、机械痛阈、热痛阈;酶联免疫吸附法检测白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超敏-C反应蛋白(hs-CRP)水平;流式细胞仪检测CD8+、CD4+水平;Western blot法检测磷酸化细胞外信号调节激酶(p-ERK)、p38 MAPK蛋白相对表达量。结果:与正常组相比,创伤组、骨髓基质干细胞组大鼠再生上皮厚度较厚,机械痛阈、热痛阈水平较低,差异有统计学意义(P<0.05)。与正常组相比,创伤组、骨髓基质干细胞组大鼠CD8+、CD4+水平较低,IL-6、TNF-α、hs-CRP水平较高(P<0.05)。与正常组相比,创伤组、骨髓基质干细胞组大鼠p-ERK、p38MAPK蛋白表达较高(P<0.05)。与创伤组相比,骨髓基质干细胞组大鼠创口愈合率较高,再生上皮厚度较厚,机械痛阈、热痛阈水平较高(P<0.05)。与创伤组相比,骨髓基质干细胞组大鼠CD8+、CD4+水平较高,IL-6、TNF-α、hs-CRP水平较低(P<0.05)。与创伤组相比,骨髓基质干细胞组大鼠p-ERK、p38MAPK蛋白表达较低(P<0.05)。结论:使用骨髓基质干细胞干预的皮肤创伤大鼠,其创口愈合率较高,再生上皮厚度较薄,炎症因子水平得到调控,免疫功能得到改善,p-ERK、p38MAPK蛋白水平得到调节,镇痛效果良好,效果显著。
Abstract:
Objective To explore the intervention effect of bone marrow stromal stem cells on skin wound repair and analgesia in rats based on p38 mitogen-activated protein kinase (p38MAPK) pathway. Methods Thirty Wistar male rats were selected, 10 were randomly selected as the normal group, and the remaining 20 rats were established as skin trauma models. Nineteen rats were successfully modeled, and they were randomly divided into trauma group (n=9) and bone marrow stromal stem cell group (n=10). After modeling, BMSCS were injected into rats in the BMSCS group at the wound site, while BMSCS in the normal group and the trauma group were not treated, and the changes of rats in each group were observed. Wound healing rate, thickness of regenerated epithelium, mechanical pain threshold and thermal pain threshold of rats in each group were compared. Enzyme-linked immunosorbent assay was used to detect IL-6, TNF-α, hs-CRP level. The levels of CD8+ and CD4+ were detected by fl ow cytometry. Western blot was used to detect the relative expression levels of p-ERK and p38 MAPK proteins. Results Compared with normal group, the thickness of regenerated epithelium was thicker in trauma group and BMSCS group, and the mechanical pain threshold and thermal pain threshold were lower ,the difference was statistically significant (P <0.05). Compared with normal group, the levels of CD8+ and CD4+ were lower in trauma group and BMSCS group, while the levels of IL-6, TNF-α and hs-CRP were higher (P<0.05). Compared with normal group, the expression of large p-ERK and p38MAPK protein was higher in trauma group and BMSCS group (P<0.05). Compared with the wound group, the wound healing rate was higher in BMSCS group, the regenerative epithelium was thicker, and the mechanical pain threshold and thermal pain threshold were higher (P<0.05). Compared with the trauma group, the levels of CD8+ and CD4+ were higher in BMSCS group, while the levels of IL-6, TNF-α and hs-CRP were lower (P<0.05). Compared with trauma group, the expression of large p-ERK and p38MAPK protein was lower in BMSCS group (P<0.05). Conclusion The wound healing rate was higher, the thickness of regenerative epithelium was thinner, the levels of inflammatory factors were regulated, the immune function was improved, the levels of p-ERK and p38MAPK protein were regulated, and the analgesic effect was good.
