摘要:
目的:探讨十精丸对于光老化大鼠皮肤朗格汉斯细胞(Langerhans cell,LCs)及体内炎症因子水平的影响。方法: SD 大鼠,雌性50只,随机分为模型组、阳性对照组、十精丸低剂量组、十精丸中剂量组、十精丸高剂量组,每组10只。建 立皮肤光老化模型,同时给予十精丸干预治疗。实验结束后,通过免疫组化法对大鼠皮肤LCs数量及形态进行观察,病理切 片观察皮肤炎性浸润程度,ELISA方法检测皮肤组织和血清中白介素-6(Interleukin-6,IL-6)、肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)含量。结果:免疫组化结果显示,与模型组相比,十精丸各剂量组表皮LCs细胞数目均有所增 加,胞体形态改善,树突结构形成增多,细胞间连接紧密。其中十精丸高剂量组改善效果较为显著。病理切片结果显示,十 精丸各剂量组大鼠皮肤炎性细胞浸润程度有所改善,十精丸高剂量组改善程度较为明显。ELISA检测结果显示,十精丸各剂 量组皮肤组织匀浆及血清中炎症因子IL-6、TNF-α含量均有所下降。其中十精丸中、高剂量组下降明显,差异有统计学意义 (P<0.01),与阳性对照组比较,差异无统计学意义(P>0.05)。结论:十精丸能增加光老化大鼠皮肤组织中LCs细胞数 量,改善LCs细胞异常形态,减轻大鼠皮肤组织的炎性细胞浸润程度,降低大鼠皮肤组织及血清中炎症因子IL-6、TNF-α的 含量,表明十精丸能提高皮肤免疫功能,抑制炎症因子,从而修复皮肤功能,改善皮肤衰老状态。
Abstract:
Objective To investigate the eff ects of Shijingwan on the levels of skin Langerhans cells (LCs) and infl ammatory cytokines in photoaged rats. Methods 50 female SD rats were randomly divided into model group, positive control group, Shijingwan low-dose group, Shijingwan medium-dose group and Shijingwan high-dose group, with 10 rats in each group. The skin photoaging model was established, and Shijingwan were given for intervention. After the experiment, the amount and morphology of LCs in the skin of rats were observed by immunohistochemistry, the degree of infl ammatory infi ltration was observed by pathological sections, and the contents of Interleukin-6 (IL-6) and Tumor necrosis factor-α(TNF-α) in the skin tissue and serum were detected by ELISA. Results Immunohistochemical results showed that, compared with model group, the number of epidermal LCs cells in each dose group of Shijingwan increased, the morphology of cell bodies improved, the formation of dendritic structure increased, and the connections between cells were tight. The improvement eff ect of Shijingwan high dose group was more signifi cant. Pathological section results showed that the degree of skin infl ammatory cell infi ltration was improved in each dose group of Shijingwan, and the degree of improvement was more obvious in the high dose group of Shijingwan. ELISA results showed that the contents of infl ammatory cytokines IL-6 and TNF-α in skin homogenate and serum of Shijingwan were decreased in each dose group. There was significant decrease in the medium and high dose groups of Shijingwan, with statistical signifi cance (P <0.01), but no signifi cant diff erence compared with positive control group (P >0.05). Conclusion Shijingwan can increase the number of LCs cells in the skin tissue of photoaging rats, improve the abnormal morphology of LCs cells, reduce the degree of infl ammatory cell infi ltration in the skin tissue of rats, and reduce the contents of infl ammatory factors IL-6 and TNF-α in the skin tissue and serum of rats, indicating that Shijingwan can improve the skin immune function, inhibit infl ammatory factors, and repair the skin function. Improve skin aging.
