Abstract:
Abstract: Objective To investigate the effect of miR-197 on proliferation and invasion of human skin squamous cell
carcinoma cell A431 by regulating JAK2. Methods Forty specimens of skin squamous cell carcinoma and forty specimens of
corresponding paracancerous tissues were collected from July 2018 to May 2019. The expressions of miR-197 in skin squamous
cell carcinoma tissue paracancerous tissue, human normal skin cell line HaCaT, human skin squamous cell carcinoma cell lines
A431, A431-miR-197-siRNA and A431-miR-197-siRNA-NC were detected by RT-qPCR. The expressions of JAK2 in various
cell lines was detected by western-Blot. The targeting relationship between miR-197 and JAK2 was verifi ed by bioinformatics
prediction and double luciferase reporter gene experiment. Effect of miR-197 on proliferation of A431 was detected by MTS.
Effect of MiR-197 on the invasive ability of skin squamous cell carcinoma cells was detected by Transwell. Effect of miR-197
on the expression of N-cadherin, a molecule related to epithelial-mesenchymal transition (EMT) was detected by IFA. Results
The expressions level of miR-197 in skin squamous cell carcinoma tissue and skin squamous cell carcinoma cell line A431
were higher than that in adjacent tissue and human normal skin cell line HaCaT respectively, the differences were statistically
signifi cant (P <0.05). Compared with A431 and A431-miR-197-siRNA-NC, the expression levels of miR-197 and JAK2 in
the stably down-regulated cell line A431-miR-197-siRNA decreased signifi cantly (P <0.05). Analysis of Target Scan database
showed that miR-197 and JAK2 had complementary binding sites. Downregulation of miR-197 could significantly reduce
the proliferation and invasion ability of skin squamous cell carcinoma cells, and inhibit the expression level of N-cadherin.
Conclusion Downregulation of miR-197 could inhibit the expression of JAK2 and reduce the expression of EMT related
molecule N-cadherin in skin squamous cell carcinoma, thus inhibiting the EMT process and proliferation and invasion ability of
skin squamous cell carcinoma cells.
