[摘要]目的:探讨miR-let-7b通过调控相关细胞周期蛋白影响皮肤黑色素瘤的增殖和凋亡。方法:qPCR法检测miR-let-7b在
黑色素瘤组织和细胞株中的表达情况;双荧光素酶报告基因检测miR-let-7b与CCND1之间的相互作用;MTT增殖实验检测抑
制miR-let-7b后黑色素瘤细胞的增殖能力的变化情况;流式细胞术检测抑制miR-let-7b后黑色素瘤细胞的凋亡行为的变化情
况。结果:与癌旁正常皮肤组织相比,黑色素瘤组织中miR-let-7b的表达水平相对上调,与其他黑色素瘤细胞株相比,A375
细胞中miR-let-7b表达最高;双荧光素酶实验证实miR-let-7b能与CCND1的3’UTR特异性结合,可以调控CCND1的表达与活
性;抑制miR-let-7b的表达后可以抑制黑色素瘤细胞的增殖能力;同时可以促进黑色素瘤细胞的凋亡行为。结论:miR-let-
7b可以调控CCND1的表达从而影响黑色素瘤细胞的增殖和凋亡。

Abstract:
Abstract: Objective To investigate the effect of miR-let-7b on the proliferation and apoptosis of melanoma by regulating
related cell cycle proteins. Methods Expression of miR-let-7b in melanoma tissues and cell lines was detected by qPCR.
Double luciferase reporter gene was used to detect the interaction between miR-let-7b and CCND1. MTT proliferation assay
was used to detect the proliferation of melanoma cells after inhibition of miR-let-7b. Flow cytometry was used to detect the
apoptotic behavior of melanoma cells after inhibition of miR-let-7b. Results Compared with adjacent normal skin tissue, the
expression level of miR-let-7b in melanoma tissue was relatively up-regulated. Compared with other melanoma cell lines, the
expression of miR-let-7b was the highest in A375 cells. Double luciferase assay confirmed that miR-let-7b could specifically
bind to the 3’ UTR of CCND1 and regulate the expression and activity of CCND1. miR-let-7b can inhibit the proliferation of
melanoma cells, And can promote the apoptosis of melanoma cells. Conclusion miR-let-7b can regulate the expression of
CCND1 and affect the proliferation and apoptosis of melanoma cells.
