[摘要]目的:本实验旨在研究正畸治疗过程中骨皮质切开术对大鼠牙龈成纤维细胞(Rat gingival fibroblasts,RGFs)的
细胞间黏附分子(intercellular adhesion molecule,ICAM)-1、血管细胞黏附分子(vascularcell adhesion molecule,
VCAM)-1表达水平的影响和作用机制。方法:选取10周雄性健康Wistar大鼠57只,随机分成骨皮质切开组、普通正畸组、骨
皮质切开+普通正畸组,每组18只;剩余3只为空白对照组。按照相应设计构建大鼠牙齿移动模型,分别于第10、20、30天处
死大鼠,提取RGFs并进行体外培养,收集各组RGFs蛋白及RNA,通过Western blot及逆转录多聚酶链式反应检测黏附分子的
蛋白表达水平及mRNA表达水平,采用SPSS 19.0软件对数据进行统计分析。结果:同一时间点(10d,20d,30d),普通正
畸组、骨皮质切开+普通正畸组RGFs的ICAM-1和VCAM-1的蛋白表达水平和mRNA表达水平均显著高于对照组,差异有统计学意
义(P <0.05);不同时间点,骨皮质切开+普通正畸组ICAM-1和VCAM-1的蛋白表达水平和mRNA表达水平随时间变化显著升高
(P <0.05)。结论:正畸治疗时采用骨皮质切开术可能诱导免疫细胞的浸润、转移并促使牙周组织炎症发生。因此,在使用
骨皮质切开术进行正畸治疗时,口腔牙周卫生维护更加重要。

Abstract:
Abstract: Objective This experiment aims to study the level effects and mechanisms process of orthodontic treatment
of bone cortex incision on intercellular adhesion molecule cell adhesion molecules(ICAM)-1 and vascular cell adhesion
molecule(VCAM)-1 expression on rat gingival fibroblast(RGF). Methods 57 healthy Wistar rats were randomly divided
into three groups: the bone cortex incision group, the orthodontic group, the bone cortex incision+orthodontic group, 18 rats
in each group. while the remaining 3 rats were the control group.Construction of rat tooth movement model according to the
corresponding design. The rats were killed in 10, 20 and 30d respectively. We extracted of RGFs and cultured in vitro. RGFs
protein and RNA were collected in each group. Detection of protein expression level and mRNA expression level of adhesion
molecules by Western blot and reverse transcriptase polymerase chain reaction. SPSS 19.0 were used the analysis. Results
At the same time point (10d, 20d, 30d), the expression levels of ICAM-1 and VCAM-1 in the orthodontic group and the bone
cortex incision+orthodontic group RGFs and mRNA were significantly higher than those in the control group, the differences
were statistically significant(P <0.05). At different time points, the expression levels of ICAM-1 and VCAM-1 in the the
bone cortex incision+orthodontic group and the expression of mRNA increased significantly with time(P <0.05). Conclusion
Bone cortex incision may induce the infiltration and metastasis of immune cells and promote the occurrence of periodontal
inflammation. Oral periodontal health maintenance is more important in orthodontic treatment with osteotomy.
