[摘要]目的:探究脂肪来源间充质干细胞(ADSCs)对快速扩张皮肤中反应挛缩的α-平滑肌肌动蛋白(α-SMA)表达的影
响,以及对成纤维细胞向肌成纤维细胞转化的影响。方法:选取健康新西兰大白兔肾周脂肪,分离提纯ADSCs并培养3代;建
立兔扩张皮肤模型,在实验组兔扩张皮肤下注射适量ADSCs悬液进行干预,对照组注射了等量的不含血清的DMEM,按照快速
扩张要求扩张;手术下获得两组兔扩张后皮肤并固定;制作切片HE染色后统计皮肤厚度,进行免疫组化染色后观察皮肤中
s-SMA阳性率;将数据进行统计学分析得出结论。结果:经ADSCs干预的扩张兔皮肤与对照组相比,皮肤挛缩率更低,扩张
皮肤的厚度较对照组厚,α-SMA阳性成纤维细胞率更低。结论:脂肪间充质干细胞可使扩张皮肤增厚,挛缩率降低,降低
α-SMA表达,抑制肌成纤维细胞形成。ADSCs可协助提高皮瓣质量。
Abstract:
Abstract: Objective To investigate the effects of adipose-derived mesenchymal stem cells (ADSCs) on the expression of α-SMA
in the contracted skin and the effect of fibroblasts on myofibroblasts. Methods The peri-renal fat of healthy New Zealand white
rabbits was taken, and ADSCs were isolated and purified for 3 generations. The model of rabbit dilated skin was established.
The rats in the experimental group were injected with appropriate amount of ADSCs suspension for intervention. The control
group was injected with the same amount of serum-free. DMEM was expanded according to the requirement of simple rapid
expansion; the skin of the two groups of rabbits was obtained and fixed under surgery; the skin thickness was counted after HE
staining, and the positive rate of s-SMA in the skin was observed after immunohistochemical staining; The analysis concludes.
Results Compared with the control group, the expanded rabbit skin treated with ADSCs had a lower skin contracture rate. The
thickness of the expanded skin was thicker than that of the control group, and the rate of α-SMA positive fibroblasts was lower.
Conclusion Adipose-derived mesenchymal stem cells can thicken the skin, reduce the contracture rate, decrease the expression
of α-smooth muscle actin and inhibit the formation of myofibroblasts. ADSCs can help improve the quality of the flap.
