摘要:
[摘要]目的:通过测定注射富血小板血浆(Platelet-rich plasma,PRP)后大鼠光老化皮肤Ⅰ、Ⅲ型胶原蛋白,转化因子β
(TGF-β),基质金属蛋白酶1(MMP-1)的表达量,观察PRP对大鼠光老化皮肤的改善作用。方法:通过紫外线照射大鼠背部
皮肤建立皮肤光老化模型。将模型动物随机分为5组,A-PRP注射组注射激活的PRP(A-PRP);N-PRP注射组注射未激活的PRP
(N-PRP);生理盐水注射组注射生理盐水;空白对照组仅照射建模;正常大鼠组仅剃除毛发。干预4周后,通过免疫组化法
对大鼠皮肤Ⅰ、Ⅲ型胶原蛋白含量进行半定量分析;通过Western bolt法对大鼠皮肤TGF-β、MMP-1表达量进行测定。结果:
Ⅰ型胶原表达:A-PRP注射组较N-PRP注射组、生理盐水注射组、空白对照组显著升高,差异有统计学意义(P <0.05)。Ⅲ
型胶原表达:A-PRP注射组显著高于生理盐水注射组、空白对照组(P <0.05),与N-PRP注射组比较,差异无统计学意义
(P >0.05)。TGF-β表达:A-PRP注射组显著高于N-PRP注射组、生理盐水注射组、空白对照组(P <0.05);MMP-1表达:
A-PRP注射组显著低于生理盐水注射组、空白对照组(P <0.05),与N-PRP注射组比较,差异无统计学意义(P >0.05)。结
论:注射PRP可以提高光老化大鼠皮肤Ⅰ、Ⅲ型胶原蛋白和TGF-β表达,降低MMP-1表达,其中A-PRP对Ⅰ型胶原和TGF-β表
达的促进作用优于N-PRP。

Abstract:
Abstract: Objective The effect of PRP on photoaging skin of mice was observed by measuring the expression of type Ⅰand
type Ⅲ collagen, TGF-β, MMP-1 in the skin of mice after PRP injection. Methods Skin photoaging model was established by
irradiating the skin of the back of rats with UV light. A-PRP injection group was injected with activated PRP (A-PRP). N-PRP
injection group was injected with inactive PRP (N-PRP). Saline injection group was injected with normal saline. Blank control
group was irradiated only with UV. Normal rat group only shave the hair. After injection for 4 weeks, the content of collagen
type Ⅰ and Ⅲ in rat skin was semi-quantitatively analyzed by immunohistochemistry. The expression of TGF-β and MMP-1 in
rat skin was determined by Western blot. Results The expression of type Ⅰ collagen in A-PRP injection group was significantly
higher than that in N-PRP injection group, saline injection group and blank control group, the differences were statistically
significant(P <0.05). The expression of type Ⅲ collagen in A-PRP injection group was significantly higher than that in group
saline injection group and blank control group(P <0.05), and there was no significant difference between A-PRP injection group
and N-PRP injection group(P >0.05). The expression of TGF-β in A-PRP injection group was significantly higher than that in
N-PRP injection group, saline injection group and blank control group(P <0.05), the expression of MMP-1 was significantly
lower in A-PRP injection group than that in saline injection group and blank control group(P <0.05). The expression of MMP-1
in A-PRP injection group and N-PRP injection group had no statistical significance(P >0.05). Conclusion Injection of PRP can
enhance the expression of type Ⅰ collagen, type Ⅲ collagen and TGF-β and decrease the expression of MMP-1 in the photoaged
rat skin. The effect of A-PRP on the expression of type Ⅰ collagen and TGF-β is better than that of N-PRP.
