[摘要]目的:观察人参皂苷Rg1抗长波紫外线(Ultravio1et A,UVA)对人皮肤成纤维细胞(Human dermal fibroblasts,HDF)
的损伤作用,为研究开发光老化防护剂提供依据。方法:分离HDF进行原代培养及冻存;复苏HDF传代培养至第6~8代用于样
品活性检测;采用改良MTT法检测UVA照射前后给予人参皂苷Rg1 HDF存活率的变化情况,用维生素C作阳性对照。结果:在照
射前24h预先给予人参皂苷单体Rg1,在2.5~40μg/ml浓度时细胞存活率较模型组明显提高,差异有统计学意义(P <0.05);
且5、10μg/ml组的细胞存活率优于阳性对照组(P <0.05)。照射后给予人参皂苷单体Rg1,在2.5~40μg/ml浓度时细胞存活
率均较模型组明显提高(P <0.05)。结论:人参皂苷Rg1在2.5~40μg/ml范围内对HDF的UVA损伤有较好的预防和治疗作用。

Abstract:
Abstract: Objective T0 observe the anti-damage effect of ginsenoside Rg1 against ultraviolet A (UVA) on human dermal
fibroblasts (HDF), and provide a basis for the research and development of photo aging protection agents. Methods Isolated
HDF for primary culture and cryopreservation. Resuscitated HDF subculture to the 6th to 8th generation for sample activity
detection. Utilizing the modified MTT method to detect the survival rate of HDF before and after UVA irradiation when
treatment with ginsenoside Rg1, and vitamin C was used as a positive control. Results The ginsenoside monomer Rg1 was
pre-administered 24h before irradiation, the cell survival rate was significantly improved compared with the model group at a
concentration of 2.5-40μg/ml, the differences were statistically significant (P <0.05). And the cell survival rate of 5, 10μg/ml
group was better than vitamin C group(P <0.05). The ginsenoside Rg1 was given after irradiation, the cell survival rate at the
concentration of 2.5-40μg/ml was significantly higher than those of the model group (P <0.05). Conclusion Ginsenoside Rg1
has a good preventive and therapeutic effect on the UVA damage ofHDF in the range of 2.5-40μg/ml.
