摘要:
目的:研究红景天苷在高海拔地区干预大鼠创伤性慢性创面后对创面愈合的影响、微血管的表达及LDH活性检测。方法:选取75只成年SD雄性大鼠,按照随机数字表法将大鼠分为3组,每组25只,并进行创伤造模。造模完成后,一般创面组只用生理盐水纱布湿敷创面,慢性创面组在此基础上连续肌注7 d氢化可的松(80 mg/kg),红景天苷组则连续肌注7 d氢化可的松(80mg/kg),并在创面湿敷溶有红景天苷的生理盐水纱布。通过分光光度计分别检测造模3、7、14 d后各组的乳酸脱氢酶(LDH)活性。HE染色及CD31免疫组织化学染色,观察第14天肉芽组织的新生毛细血管分布情况及微血管密度。第7、14、21、28d观察各组创面愈合情况,计算创面愈合率。结果:红景天苷组LDH活性在造模后3、7、14d均低于一般创面组及慢性创面组,差异均具有统计学意义(P<0.05)。第14天时,红景天苷组HE染色下创面新生毛细血管,CD31免疫组织化学染色下CD31表达及微血管密度均高于一般创面组及慢性创面组(P<0.05)。红景天苷组创面愈合率明显高于一般创面组及慢性创面组(P<0.05),红景天苷组创面愈合情况最好,皮肤边缘纤维组织增生最少,创面未见明显渗出及炎症反应。结论:红景天苷干预可促进大鼠创伤性慢性创面的愈合,促进微血管生成,减轻组织损伤,加速创面愈合。
Abstract:
Objective To study the effect of salidroside on wound healing, microvascular expression and LDH activity after intervening in traumatic chronic wound in rats at high altitude. Methods Forty five SD male rats were selected to make trauma models.The rats were divided into 3 groups according to the random number table method,which were general wound group,chronic wound group and salidroside group.In the general wound group, only normal saline gauze was used to wet compress the wound, in the chronic wound group, hydrocortisone(80mg/kg) was injected intramuscularly for 7 days,and in salidroside group, hydrocortisone (80 mg/kg) was injected intramuscularly for 7 days, and salidroside dissolved normal saline gauze was wet applied to the wound.The activity of lactate dehydrogenase (LDH) in each group was detected by spectrophotometer in 3rd, 7th, 14th day.Hematoxylin eosin staining and CD31 immunohistochemical staining were performed to observe the distribution of new capillaries and microvessel density in granulation tissue on 14th day . The wound healing of each group was observed on the 7th,14th,21st,28th day to observation and the wound healing rate was calculated. Results LDH activity in salidroside group was lower than that in general wound group and chronic wound group at 3rd, 7th and 14th days,the comparative differences were statistically significant (P<0.05). On the 14th day, the expression of CD31 and microvessel density in salidroside group were higher than those in general wound group and chronic wound group,the comparative differences were statistically significant(P<0.05). The wound healing rate of salidroside group was significantly higher than that of general wound group and chronic wound group,the comparative differences were statistically significant(P<0.05). The wound healing in salidroside group was the best, the proliferation of fibrous tissue on the edge of the skin was the least, and there was no obvious exudation and inflammatory reaction on the wound. Conclusion The intervention of salidroside can promote the healing of chronic traumatic wounds in rats, promote microangiogenesis, reduce tissue damage, and accelerate wound healing
