[摘要]目的:探讨不同大小的动态载荷下人牙周膜细胞(hPDLCs)中线粒体发育和功能水平的变化。方法:体外分离因正畸
治疗需要而拔除的健康前磨牙的牙周膜组织,培养健康的hPDLCs。利用动态流体静压加力装置加载0~50kPa、0~90kPa、
0~150kPa的周期性动态压应力1d,3d,5d。采用MTT检测其对细胞增殖活性的影响,采用实时定量PCR分别检测线粒体发育及
功能相关基因的RNA表达水平。结果:与未受力组细胞相比,0~50kPa、0~90kPa组hPDLCs中线粒体发育相关基因PGC-1α、
TFAM、NRF1及线粒体功能相关基因Cox7a1、Cox8b RNA表达水平增加,而0~150kPa组hPDLCs中以上线粒体发育及功能相关基
因RNA表达水平则显著降低。结论:适当的机械应力刺激条件能增加人牙周膜细胞中线粒体发育水平并促进其功能,而过大
载荷则会抑制线粒体的发育及生理功能的发挥。

Abstract:
Abstract: Objective To investigate the changes of mitochondrial development and function in human periodontal ligament
cells (hPDLCs) under different dynamic loads. Methods The periodontal ligament tissues of healthy premolars extracted
for orthodontic treatment were isolated in vitro, and healthy hPDLCs were cultured. Cyclic dynamic compressive stresses
of 0-50kPa, 0-90kPa and 0-150kPa were loaded by dynamic hydrostatic pressure device for 1, 3 and 5 days. MTT was used
to detect its effect on cell activity, and real-time quantitative PCR was used to detect the expression level of mitochondrial
development and function-related genes. Results Compared with the cells in the non-stressed group, the expression levels
of mitochondrial development genes PGC-1α, TFAM, NRF1 and mitochondrial function-related genes Cox7a1 and Cox8b
increased in the 0-50kPa and 0-90kPa groups, while the expression levels of mitochondrial development and function-related
genes decreased significantly in the 0-150kPa groups. Conclusion Proper mechanical stress stimulation can increase the level
of mitochondrial development and promote its function in human periodontal ligament cells, while excessive loading can inhibit
the development and physiological function of mitochondria.
