摘要:
[摘要]目的:研究萝卜硫素对中波紫外线(UVB)照射后黑素细胞产生黑素能力的抑制效应及其对Traf6/TAK1信号传导的影
响。方法:采用人表皮黑素PIG1细胞为研究模型,分为对照组(Control)、模型组(Model,UVB照射)、萝卜硫素处理
组(10、20、40μM)。CCK8法分析PIG1细胞增殖情况,氢氧化钠裂解法分析黑素含量,酶联免疫吸附试验(ELISA)分析
优黑素含量,Western blot技术检测小眼畸形相关转录因子(MITF)、酪氨酸酶(TYR)、酪氨酸酶相关蛋白-1(TRP-1)、
TRP-2、肿瘤坏死因子相关受体因子6(Traf6)及转录生长因子β-活化激酶1(TAK1)表达。结果:与对照组比较,UVB照射
能诱导PIG1细胞黑素及优黑素含量显著升高(P<0.05),还能促使MITF、TYR、TRP-1、TRP-2、Traf6及p-TAK1蛋白表达水平
显著增高(P <0.05);与模型组比较,不同浓度萝卜硫素能降低PIG1细胞黑素及优黑素的生成(P <0.05),还能显著抑制
MITF、TYR、TRP-1、TRP-2、Traf6及p-TAK1蛋白表达(P <0.05),呈现出剂量依赖性。结论:萝卜硫素通过抑制Traf6/TAK1
信号传导降低UVB诱导PIG1细胞黑素生成。

Abstract:
Abstract: Objective To study the suppressed effects of sulforaphane on ultraviolet radiation B( UVB)-stimulated melanogenesis of
melanocytes(PIG1), and observe its effect on Traf6/TAK1 signal pathway. Methods PIG1 cells were cultured in vitro and divided into
control group, UVB group(Model group), sulforaphane intervention groups(10,20 and 40μM). The cell proliferation was determined
by CKK8 assay. Melanin and eumelanin were detected by NaOH splitting and ELISA assay, respectively. The expression levels of
microphthalmia-associated transcription factor(MITF), tyrosinase(TYR), tyrosinase-related protein-1(TRP-1), TRP-2, tumor necrosis
factor associated receptor factor 6(Traf6), transforming growth factor-β activated kinase-1(TAK1) were measured by Western blot.
Results Compared with control group, the contents of melanin and eumelanin were significantly increased(P <0.05); the
expression levels of MITF, TYR, TRP-1, TRP-2, Traf6 and p-TAK1 in PIG1 cells were also significantly increased(P <0.05).
Compared with model group, the expression levels of MITF, TYR, TRP-1, TRP-2, Traf6 and p-TAK1 were significantly
suppressed, and production of melanin and eumelanin reduced in a dose dependent manner in each sulforaphane intervention
group(P <0.05). Conclusion Sulforaphane inhibits melanin and eumelanin production through down-regulating Traf6/TAK1
signal pathway in UVB-irradiated melanocytes.
