摘要:
[摘要]目的：通过体外细胞学研究（共培养体系建立）直观表现脂肪干细胞（Adipose derived stem cells，ADSCs）体外
进血管形成能力，阐述脂肪干细胞作为辅助细胞通过促进早期血供建立而提高移植脂肪存活率的可能机制。方法：临床通过
Coleman法抽吸成人大腿或腹部浅层脂肪，并通过经典方法培养获取ADSCs。对ADSCs进行形态学、表面分子标记物及多向分
化等生物学特性鉴定，并通过Matrigel培养脐静脉内皮细胞（Human umbilical vein endothelial cells，HUVECs）管腔
样结构形成实验探讨ADSCs体外促血管形成能力，说明ADSCs促进血管新生及移植物早期血供建立的潜力。结果：ADSCs/
HUVECs共培养体系可于体外促进内皮细胞形成血管腔样结构，提高早期血管新生。结论：通过共培养体系模拟提示ADSCs
体外具备促血管形成的能力，具有促进早期血供建立而提高移植物存活的潜力，为临床细胞辅助脂肪移植提高存活率的可
能机制。

Abstract:

Abstract: Objective To evaluate the angiogenesis potential of the adipose derived stem cells (ADSCs) in vitro in order to
expound the possible mechanism about the ADSCs improved the survival rate of graft through promoting vascular remodeling
in early stage. Methods The fat tissue was obtained from healthy adult’s abdomen or thigh liposuction through the Coleman
technique. The ADSCs was obtained and cultured by the traditional method. The ADSCs was Identified by biological function
properties studies including morphological characteristics, molecular surface marker expression and differentiation. The
angiogenesis potential was testified by the tube formation assay system in Matrigel with Human umbilical vein endothelial
cells (HUVECs) in order to expound the angiogenesis potential for improving the survival rate of graft. Results ADSCs
promoted the tube formation in vitro in the co-culture system in order to promote the angiogenesis in early stage. Conclusion
By co-culture system, the ADSCs had the ability to promote angiogenesis. It had the potential that promote establishing the
blood supply at early stage in vitro to improve the survival rate of graft, and that is the plausible mechanisms for cell assisted
lipotransfer to improve the survival rate.
