摘要:
目的:观察丹参酮ⅡA对大鼠环孢素A(CyclosporinA,CsA)性牙龈增生上皮内Bcl-2表达的影响。方法:30只7周龄雄性Wistar大鼠随机等分为3组(对照组、CsA组、CsA+TanⅡA组),每组再随机等分为2个亚组(10 d组和20 d组)。实验周期结束时取材并制作切片,选择相同部位分别行HE染色及免疫组化PV两步法,于显微镜下观察并测量各组大鼠颊侧牙龈上皮面积和最长钉突长度,计数最长钉突内Bcl-2蛋白阳性细胞表达数,行完全随机分组两因素析因设计的方差分析。结果:颊侧牙龈上皮面积、最长钉突长度、抗凋亡蛋白Bcl-2阳性细胞数,CsA组显著大于对照组和CsA+TanⅡA组(P<0.05);而CsA+TanⅡA组和对照组相比,差异无统计学意义(P>0.05)。结论:在本试验周期内,TanⅡA能有效地预防CsA引起的大鼠牙龈上皮增厚,Tan ⅡA抑制大鼠CsA性牙龈增厚上皮内抗凋亡蛋白Bcl-2的表达可能是其机制之一。
Abstract:
Objective This paper is to observe the eff ect of Tanshinone ⅡA (TanshinoneⅡA, TanⅡA) on the expression of Bcl-2 in the epithelium Cyclosporin A (CyclosporinA, CsA)-induced gingival overgrowth of rats. Methods Thirty 7-week-old male Wistar rats were randomly divided into 3 groups (control, CsA, CsA+TanⅡA), and each group was randomly divided into 2 subgroups (10 days, 20 days). At the end of the experimental cycle, samples were taken and sectioned, and the same parts were selected for HE staining and immunohistochemical PV. The area of buccal gingival epithelium and the length of the longest nail process of rats in each group were observed and measured under the microscope, the imaging analysis system was used to take pictures and measure the buccal gingival epithelial area and the longest spike length of each group of rats, count the number of Bcl-2 protein positive cells in the longest spike, and analyze two-way analysis of varianca of factorial design. Results The area of buccal gingival epithelium, the length of the longest nail process and the number of anti apoptotic protein Bcl-2 positive cells in CsA group were signifi cantly higher than those in control group and CsA + Tan ⅡA group (P<0.05), there was no signifi cant diff erence between CSA + Tan ⅡA group and the control group (P>0.05). Conclusion Within this test period, Tan ⅡA can eff ectively prevent CsA-induced gingival epithelial overgrowth in rats. Tan ⅡA inhibits the expression of intraepithelial antiapoptotic protein Bcl-2 in CsA-induced gingival overgrowth in rats may be one of its mechanisms
