[摘要]目的:探讨维生素K3对UVA辐照下角质形成细胞的光保护作用及相关机制。方法:以5J/cm2的UVA辐照HaCaT细胞建立光
老化细胞模型。采用不同浓度(0、0.125μM、0.25μM、0.5μM)的维生素K3预处理HaCaT细胞24h;CCK-8方法检测细胞增
殖活性;活性氧(ROS)检测荧光探针2’,7’-二氯荧光黄双乙酸盐(DCFH-DA)检测细胞内活性氧水平;Western blot法检测
各组细胞MMP1、COX2、LC3B蛋白的表达。结果:与单纯光照组相比,维生素K3能够促进光老化HaCaT细胞的体外增殖,且呈
浓度依赖性,差异有统计学意义(P <0.05);药物干预组HaCaT细胞的ROS含量降低,差异有统计学意义(P <0.05);细胞
内MMP1、COX2蛋白表达降低,LC3B表达增高,差异有统计学意义(P <0.05)。结论:维生素K3能够对UVA辐照HaCaT细胞提
供光保护作用,其机制可能与促进细胞自噬有关。

Abstract:
Abstract: Objective To explore the photoprotective effect of Vit K3 on keratinocytes under UVA irradiation and related
mechanisms. Methods HaCaT cells were irradiated with 5J/cm2 of UVA to establish a photoaging cell model. HaCaT cells
were pretreated with Vit K3 at different concentrations (0, 0.125μM, 0.25μM, 0.5μM) for 24h. Proliferation of cells was detected
by CCK-8 method. Reactive oxygen species (ROS) detection fluorescent probe 2’,7’-dichlorofluorescein yellow diacetate
(DCFH-DA) was used to detect intracellular reactive oxygen level. The expression of MMP1, COX2 and LC3B was detected
by Western blot. Results Compared with the light-only group, Vit K3 could promote the proliferation of HaCaT cells in a
concentration dependent manner, the difference was statistically significant (P <0.05). The ROS content of HaCaT cells in the
drug intervention group was lower (P <0.05). The expression of MMP1 and COX2 protein was decreased, and the expression
of LC3B was increased (P <0.05). Conclusion Vit K3 could provide photoprotection to HaCaT cells irradiated by UVA, and its
mechanism might be related to promoting autophagy.
