摘要:
目的:探讨脂肪干细胞(Adipose derived stem cells,ADSCs)与放射损伤成纤维细胞(Fibroblast,Fb)共培养 后细胞因子表达的差异。方法:使用剂量8 Gy的X线放射源,对大鼠的成纤维细胞进行单次照射,建立ADSCs与放射后成纤 维细胞共培养模型。分别把未干预的成纤维细胞设为空白对照Fb1组,放射后成纤维细胞设为Fb2组,ADSCs与放射后成纤 维细胞共培养组设为Fb3组,利用蛋白质芯片检测各组差异表达的细胞因子,并通过Elisa检测验证。结果:研究结果发现 Fb2 组粒细胞-巨噬细胞集落刺激因子(Granulocyte-macrophage colony stimulating factor,GM-CSF)的表达水平低于 Fb1 组(P<0.05),Fb3组GM-CSF表达水平高于Fb2(P<0.01);Fb3组血管内皮生长因子(Vascular endothelial growth factor,VEGF)表达水平高于Fb2组(P<0.05);各组表皮细胞因子(Epidermal growth factor,EGF)和血小板衍生因子 (Platelet derived growth factor,PDGF)含量差异无统计学意义。结论:ADSCs与放射后成纤维细胞共培养后多种生长 因子、黏附因子、趋化因子上调,部分炎性因子下调;ADSCs可能主要通过VEGF和GM-CSF促进成纤维细胞放射损伤的修复。
Abstract:
Objective To investigate the diff erence in cytokines expression after co-culture of ADSCs and radiation-damaged fi broblasts. Methods An X-ray radiation source of 8 Gy was used to illuminate rat fi broblasts to establish a co-culture model of ADSCs and radiation-damaged fi broblasts. Unintervened fi broblasts were set as blank control Fb 1,radiation damaged fi broblasts as Fb2, and ADSCs with radio-injured fi broblasts as Fb3. Diff erentially expressed cytokines were detected in each group using protein microarray, which were verified by Elisa. Results The expression of granulocyte-macrophage colony stimulating factor(GM-CSF) in Fb2 group were lower than that in Fb1 group (P<0.05),while the expression of GM-CSF in Fb3 group were higher than that in Fb2 group (P<0.01); Moreover,the expression of vascular endothelial growth factor(VEGF) in Fb3 group were higher than that in Fb2 group (P <0.05). However, there was no signifi cant diff erence in epidermal growth factor(EGF) and platelet derived growth factor(PDGF) content in each group. Conclusion Some growth factors, adhesion factors, and chemokines were up-regulated while some infl ammatory factors were down-regulated after co-culture of ADSCs with radiation damaged fi broblasts. ADSCs may promote the repair of fi broblast radiation damage mainly through VEGF and GM-CSF.
