[摘要]目的:通过体外培养人乳腺癌难愈合创面中成纤维细胞,并应用TGF-β/Smad信号通路阻断剂干扰素γ(IFN-γ)干
扰后,探讨IFN-γ在乳腺癌难愈合创面中所发挥的作用和影响。方法:采用酶消化法对人乳腺癌溃疡创面组织行体外成纤维
细胞原代培养。将实验细胞分成A、B、C、D共四组,A组为IFN-γ 5 000U/ml组;B组为IFN-γ 10 000U/ml组;C组为对照
组,不加药物;D组为空白组。分别于第7天、第14天将培养细胞接种于96孔板内,用MTT法测定细胞增殖情况。结果:第7
天及第14天A组吸光值与对照组C组比较差异无统计学意义(P >0.05);B组吸光值均低于对照组C组,差异具有统计学意
义(P <0.05)。结论:IFN-γ对成纤维细胞的增殖抑制受到浓度的影响,浓度10 000U/ml时明显抑制了体外培养成纤维细
胞的增殖,而在较低浓度(5 000U/ml)时对体外培养成纤维细胞增殖无明显抑制作用。

Abstract:
Abstract: Objective The role and influence of IFN-γ in refractory wounds of breast cancer were investigated by in vitro
culture of fibroblast cells in refractory wounds of breast cancer and interference with IFN-γ (TGF-β/Smad signaling pathway
blocker). Methods In vitro fibroblast primary culture was performed on the wound tissue of human breast cancer ulcer by
enzyme digestion. The experimental cells were divided into four groups: group A, group B, group C and group D. Group A
was IFN-γ 5 000U/ml group, group B was IFN-γ 10 000U/ml group; group C was the control group without drugs, group
D was blank group. The cultured cells were inoculated into 96-well plates on the 7th and 14th day respectively, and cell
proliferation was determined by MTT method. Results There was no significant difference in OD value between group
A and control group C on the 7th and 14th day(P >0.05). The OD value of group B was lower than those of group C, the
differences were statistically significant(P <0.05). Conclusion The inhibition of IFN-γ on proliferation of fibroblasts was
affected by the concentration, the concentration at 10 000U/ml significantly inhibited proliferation of fibroblasts cultured in
vitro, while the lower concentration of IFN-γ at 5 000U/ml no obvious inhibitory effect on the proliferation of fibroblasts.
